In vivo study | GS-4997 are orally biologically active. After oral administration, GS-4997 targets and binds to the catalytic kinase domain of ASK1 in an ATP-competitive manner, inhibiting its phosphorylation and activation. GS-4997 inhibit the phosphorylation of ASK1 downstream kinases, such as c-Jun N-terminal kinases (JNKs) and p38 MAPK. By inhibiting the activation of ask1-dependent signal transduction pathway, GS-4997 can prevent the production of inflammatory cytokines, down-regulate the expression of genes involved in the process of fibrosis, inhibit excessive apoptosis and cell proliferation. |
